Abstract
The use of a gradient in HPLC analysis allows for a more complete separation of complex mixtures of ergot alkaloids, and the use of fluorescence detection permits sensitive and specific detection of those compounds having a D-8,9 configuration in the ergoline ring. This configuration also permits an epimer to form spontaneously at asymmetric C-8, which can be used to confirm the identity of analytes in complex mixtures. In the analysis of endophyte infected (EI) plants and seeds, many fluorescent peaks are commonly observed. The purpose of this study was to develop an analytical method which would resolve all of the peaks extracted from EI materials, and to identify these peaks by epimerization and mass spectrometry.
