Abstract
The signal transduction mediating PAF-induced MAPK activation in CHO-cells stably expressing cloned PAF-receptor, has been extensively studied in our group (1). Pertussis toxin (PTX) blocked about 50% of the PAF-induced MAPK activation, without altering phosphatidylinositol (PI) hydrolysis and calcium responses. It was therefore speculated that the PAF-receptor mediates MAPK activation through one PTX-insensitive pathway that yields PI hydrolysis and possibly activation of conventional protein kinase C (cPKC), and another PTX-sensitive pathway that is likely to be independent of calcium and cPKC but mainly remains obscure.
