The Controlled Release of a 5-Fluorouracil Prodrug from a Biodegradable Matrix

Abstract

A prodrug of 5-fluorouracil [5-FU] was prepared by reacting a 5-FU slurry with hexylisocyanate, in dioxane, at room temperature. The resulting solid, called HIFU, was purified and the release rates of 5-FU were determined, in distilled water, and compared against 5-FU using the gas dispersion tube technique previously described, at both room temperature and at 37°C. HIFU was studied in the form of a powder and as a monolithic dispersion in poly(caprolactone), The prodrug releases 5-FU very rapidly in the powdered form and both HIFU and 5-FU gave the typical monolithic release profiles, which were linear when plotted against the square root of time.
