Analysis of the Actions of Anti-Inflammatory Drugs, with Particular Reference to Effects on Interleukins and Leucocytes

The importance of IL’s* and other cytokines in chronic inflammation has aroused interest in their assay in contexts such as inflammatory exudates and stimulated or unstimulated leucocytes. Assays developed for two main IL’s, viz. IL-1 and IL-2, include cell responses such as production of other mediators, or reactions (e.g. proliferation, aggregation) whose specificity depends on the responsiveness of the cell type as well as the purity of the extracts, fluids or culture media being assayed, and on the assay conditions. Certain cell clones are more selective in the response to the IL’s (e.g. the NOB-1 sub-clone of the EL-4 cell for IL-1), so improving the specificity of the LAF assay. Conditions are described for improving the specificity and responses of IL-1 in the CRA using the bovine nasal septum. The recent development of RIA’s and ELISA’s for assaying IL-1 and IL-2 is considered together with limitations and conditions in applying the methods. Appropriate animal models for studies of the effects of anti-rheumatic agents are discussed in relation to the use of the above-mentioned assays.
