Comparison of an Enzyme Method with a Capillary GC Method for Red Blood Cell Sorbitol Assay

The enzyme aldolase reductase is believed to play a crucial role in the long-term complications of diabetes. This enzyme converts glucose to sorbitol and is present in several tissues and also in erythrocytes. At elevated glucose levels (as in diabetes) the pathway is activated and sorbitol accumulates. The determination of erythrocyte sorbitol thus provides a convenient indicator of aldose reductase activity. Several methods are capable of measuring sorbitol including the enzyme sorbitol dehydrogenase (SDH) [1], HPLC [2], both packed-[3] and capillary-column [4] GC; all have been tried in this laboratory.
