Analysis of eicosanoids in biological matrices is difficult. Their levels are very low, and they are rapidly metabolized to numerous structurally similar substances often found in much higher concentration. Hence past assays of insufficient specificity have given erroneous values. GC-MS, which is more specific than RIA, is effective for structure elucidation and assay of eicosanoids and metabolites, yet may be insufficiently selective at low eicosanoia concentrations. PGE2 and PGF of renal origin are present in human urine, as now shown also for PGD2 by GC-MS/MS of the methoxime-TMS* ether of its methyl ester.
