Immunoassay of Beclomethasone 17,21-Dipropionate and Metabolites

A radioimmunoassay (RIA) has been developed for determining beclomethasone 17,21-dipropionate (BDP) in human body fluids, after solvent extraction, at levels down to 0.1 (plasma) or 1 (urine) ng/ml. The 3-O-carboxymethyloxime derivative of BDP was coupled by a mixed-anhydride reaction to bovine serum albumin (BSA) for raising a rabbit antiserum, or to [125 I] iodohistamine to provide a radiolabel. Known BDP metabolites and endogenous steroids had negligible cross-reactivity.
