Abstract

In this chapter we will survey the applications of video to various modes of light microscopy including low-light-level applications, contrast enhancement, quantitation, scanning, and three-dimensional reconstruction. Rather than limit the discussion to applications that have been published, I shall also include some ongoing developments and potential applications of video that could further improve the image or otherwise expand the utility of the light microscope. Some recent applications of video to biological light microscopy are reviewed comprehensively by Spring (1979, 1983) (see also footno te ‡ on page 397 and the Postscript).
