Abstract
The overall objective is to determine the conditions under which two immobilized enzymes acting in sequence are most efficient. Consider the following sequential reaction (also called a “coupled” reaction),
with El and E2 being enzymes. We define an efficient system as one in which a minimal amount of E2 to El approximates the maximum rate obtained when E2 is in excess. The practical objectives are to utilize this type of system, or extensions of it, to either generate NAD(P)H for synthetic processes, or to recycle the pyridine nucleotide coenzyme in both synthetic and diagnostic procedures.
