Abstract
Arachidonic acid released from membrane phospholipids undergoes oxygenation by two distinct pathways that differ in terms of both their products and their susceptibility to regulation by biological and pharmacological agents(Fig. 1). The cyclooxygenase pathway yields unstable endoperoxide intermediates that undergo spontaneous or enzymatic conversion to a variety of products including pro staglandins, pro st acyclin , and thromboxanes. Oxygenation of arachidonic acid by the lipoxygenase pathway generates a series of unstable hydroperoxy-eicosatetraenoic acids (OOHETES or HPETES), of which 5-OOHETE is quantitatively predominant in several types of leukocytes, 12-OOHETE is the major product in platelets, and 11-OOHETE and 5-OOHETE in rabbit alveolar macrophages (Borgeat and Samuelsson, 1979a; Goetzl and Sun, 1979; Valone et al., 1980).
