Abstract

This chapter is intended for a later stage in purification efforts, when a method has been established or published method has been tried but there may be some dissatisfaction with the overall procedure. The objectives of a scheme for the purification of an enzyme or other protein may be any or all of the following:

    High overall recovery of activity

  2. 2.

    High degree of purity of the end product

  3. 3.

    Reproducibility

    1. (i)

      within the laboratory

    2. (ii)

      if scaled up or scaled down

    3. (iii)

      in other laboratories

  4. 4.

    Economical use of reagents and simple apparatus

  5. 5.

    Convenience in terms of working hours.
