P-450 Cytochromes in the Human Placenta: Oxidations of Xenobiotics and Endogenous Steroids

Abstract

The purpose of this article is to provide the reader with a concise view of current concepts concerning the family of P-450 hemoproteins as they exist and function in the human placenta. The subject is discussed with focus on the monooxygenation of small, foreign organic molecules (xenobiotics) in placental tissues and cells, and of the relationships of these monooxygenation reactions to placental P-450-dependent oxidations of endogenous steroids. Several past reviews of placental biotransformation of xenobiotics have appeared in the open literature and references to these are provided for readers more seriously interested in this highly specialized topic. The first review of this research area appeared in 1969 (Juchau and Yaffe, 1969). A very comprehensive and detailed review of the earlier literature appeared in 1973 (Juchau, 1973). A more recent, comprehensive, and readily accessible review appeared in 1980 (Juchau, 1980) and the latest review is in press at the time of this writing (Juchau and Rettie, 1985). Dr. Olvai Pelkonen of Finland has actively researched this area and has also provided relatively recent reviews (Pelkonen, 1980; Pelkonen and Pasanen, 1984). Other reviews appearing between 1969 and 1985 are referenced in those cited above.
