In Vitro Studies of Photochemically Cross-Linked Protein-Nucleic Acid Complexes. Determination of Cross-Linked Regions and Structural Relationships in Specific Complexes

  • Chapter

Abstract

It is well known that many enzymatic and cellular processes involve specific protein-nucleic acid associations. Examples include the interaction of a repressor with its operator DNA (see Beckwith and Zipser, 1970; Müller-Hill, 1971), of DNA polymerase with DNA (see Kornberg, 1975), of ribosomal proteins with ribosomal RNA (see Nomura et al., 1974), of aminoacyl tRNA synthetases with tRNAs (see Kisselev and Favorova, 1974; Söll and Schimmel, 1974), as well as many other systems. A prime goal is to determine the mechanism by which these interactions occur; viz., it is desirable to understand how specificity is achieved and to determine what factors are responsible for the strength of specific protein-nucleic acid associations.
