Analysis of Antigen-Specific T Cell-Mediated Suppression at the Clonal Level

For the purpose of investigating inhibition of T cell activation at a molecular level, we have assembled a panel of T cell hybridomas whose members respond to antigen and Class II molecules by production of the cytokine, IL-2, and have established bulk cultured Ts cells which inactivate certain T hybridomas. Several important observations concerning the interaction between T hybridomas and Ts cells have contributed to our understanding of inhibitory cellular interactions. These findings are 1) antigen-induced cytokine production by HEL/I-Ab specific T hybridomas is specifically and completely inhibited by HEL-induced Ts cells, 2) T cell hybridomas of other specificities (e.g. HEL/I-Ad) are not inhibited by HEL-Ts cells, 3) bulk cultured Ts cells are functionally superior to primary Ts cells following their short term culture in lymphokine supplemented media. This experimental system now provides an excellent opportunity to learn how helper T cells become inactivated by Ts and to describe the mechanism of inactivation in molecular terms.
