Monoclonal Antibodies to Cell Surface Antigens on Lymphoid and Non-Lymphoid Cells of the Mouse

Abstract

The structural basis of the complex micro-architecture of lymphoid microenvironments is still poorly understood. Further insight into the structure and function of various lymphoid microenvironments, and their role in the differentiation of subpopulations of lymphocytes can be obtained when the distribution of cell-type specific surface markers is included in histological studies. We have previously employed monoclonal antibodies directed against H-2 antigens, Thy-1, T-200 and Lyt antigens to analyze lymphoid microenvironments in frozen sections of the thymus and peripheral lymphoid organs (Rouse et al., 1979; Van Ewijk et al., 1980a, 1980b, 1981). In the present paper we describe the reactivities of five monoclonal antibodies, recently produced in our own laboratory to mouse (C3H) thymocytes (Van Vliet et al., 1980) and we compare these reactivities with well defined monoclonal anti-Thy-1, anti-Lyt-1 and anti-Lyt-2 antisera. To this purpose we have used an ELISA developed by Lansdorp et al. (1980), immunohistology and flow cytofluorometric analysis.
