Summary
We studied the effect of denervation on the extracellular matrix of a newt limb regenerate with a specific dye of collagen, Sirius red. Blastemata harvested on adult animals 4 days after denervation accumulated more collagen (+54%) than blastemata harvested just after denervation. On the other hand, no difference appeared when denervation was performed on young animals. The distribution of collagens of types I, III and IV was analyzed before and after denervation using immuno-cytochemical techniques. The results showed that the deposition of collagen in denervated blastemata concerned types I and IV. The basement membrane zone was considerably thicker in denervated regenerates. We hypothesize that the basement membrane that takes place after denervation could constitute a barrier to epidermal proteolytic enzymes and would result in an enlarged collagen deposition between mesodermal cells.
