Prevention and Treatment of Friend Leukemia Virus (FLV) Infection by Interferon-Inducing Synthetic Polyanions

Abstract

Divinyl ether maleic anhydride (MA) copolymers (pyran copolymer, NSC 46015) inhibits Friend leukemia virus (FLV) splenomegaly. Pyran copolymer induces interferon in normal mice and the decreases in splenic FLV titers seen following pretreatment with these polyanions might be explained by the appearance of interferon. In vivo inhibition of FLV splenomegaly has also been obtained by statolon, an anionic polysaccharide, and the plant lectin, phytohemagglutinin, both of which have been reported to induce interferon. Similar inhibition of FLV splenomegaly has also been seen for methyl vinyl ether/MA and styrene/MA copolymers. Pyran copolymer is currently undergoing clinical trial in man. Its animal and human pharmacology indicates uptake in the nucleated formed elements of the blood, and stimulation of reticuloendothelial activity in liver, spleen, and bone marrow.
