Differential P21 Expression and Point Mutations of ras Gene Family in Human Carcinoma Tissues

Abstract

The mammalian ras oncogene family consisits of three well-characterized members, N-ras, K-ras and H-ras. The amino acid sequence of the 21-Kd proteins encoded by these three ras members is known to be nearly identical except in two short variable regions1,2. Concerning the expression of ras p21 in human tissues, many immunohistochemical studies have been reported using monoclonal antibodies against ras protein3–5. However, these antibodies were unable to distinguish N-ras, K-ras and H-ras p21, respectively. Recently, monoclonal antibodies against each of N-ras, K-ras and H-ras p21 have been developed and made available commercially. In the present study, we examined the differential p21 expression of the three members of ras family in human carcinoma tissues of various organs by the immunohistochemical method.
