Thermo-Responsive Polymeric Hydrogels and Their Application to Pulsatile Drug Release

Abstract

Poly(N-isopropyl acrylamide)(IPAAm) has characteristics to exhibit a remarkable swelling change in aqueous system in response to step-wise temperature change. Random copolymers of IPAAm with alkyl methacrylate (RMA) having different length of alkyl side chain were synthesized to achieve complete on-off drug permeation and release in response to step-wise temperature. On-off release and permeation of indomethacin were found to be due to surface shrinking layer with increasing temperature. The formation process of these surface shrinking layers were confirmed to be controlled by alkyl side chain length of RMA. From these results, the effects of chemical structure (length of alkyl side chain) on temperature modulated drug permeation and palsatile drug release pattern have been discussed.
