Abstract
Blood samples were obtained from 61 volunteers working in a Finnish iron foundry who were occupationally exposed to polycyclic aromatic hydrocarbons (PAHs) from 19 control subjects not known to be occupationally exposed to this class of chemical carcinogens. Foundry workers were classified as belonging to high, medium, or low exposure groups according to their exposure to airborne benzo(a)pyrene (BaP) (high > 0.2, medium 0.05–0.2, low < 0.05 µg BaP/m3 air). Aromatic adducts were determined in white blood cell DNA from the subjects using the 32P-postlabeling technique. There was a high correlation between the estimated exposure and adduct levels.
Differences in the level of adducts were also noted between job categories in the foundry. This study suggests that the 32P-postlabeling assay may be useful in monitoring human exposure to known and previously unidentified environmental genotoxic agents.
