Pulsatile Insulin Release and Electrical Activity from Single ob/ob Mouse Islets of Langerhans

Abstract

Since Ca-action potentials were first measured in β-cells, it has been assumed that the resultant Ca2+ influx provides a critical Ca2+-dependent step in the events leading to insulin secretion.4,6. The correlation between electrical activity and insulin release remained indirect, however, since membrane potentials were recorded from single β-cells within an isolated islet and insulin release was measured from batches of collagenase isolated islets (usually more than 100) or from the perfused pancreas (containing over 1000 islets).
