Abstract

Most strategies used to isolate genes from recombinant DNA libraries have employed nucleic acid probes (1,2). Genes can also be identified and isolated through the use of antibody probes of the antigen(s) expressed by specific genes (3). This manuscript reviews the observations which led to the development of a system for efficient gene isolation using antibody probes, includes protocols for constructing and screening λgtll recombinant DNA expression libraries and describes strategies which can be used to confirm gene identity.
