Abstract

Growth of the yeast Rhodotorula graminis on benzoate results in the induction of benzoate hydroxylase, p-hydroxybenzoate-3-hydroxy-lase, protocatechuate-3,4-dioxygenase, and enzymes of the ß-ketoadi-pate pathway. Following subcellular fractionation of crude extracts of benzoate-grown cells, benzoate hydroxylase activity was observed in the particulate (membrane) fraction. Thus, washed membrane preparations, which are devoid of other benzoate-dissimilatory enzymes, were used as a source of partially purified benzoate hydroxylase for further study.
