Abstract
The hemoflagellates that infect man have proven recalcitrant to effective chemotherapy. The specificity of antimicrobial therapy, so useful in the treatment of diseases caused by bacteria, is lacking in diseases caused by these protozoans which have adapted themselves to survival within mammalian cells. This is evinced by the significant toxicity of the agents used in their treatment. A high therapeutic index for a given antimicrobial usually requires a specific difference in metabolism between the host and the parasite. Such a difference appears to exist in the ability of organisms of the genera Leishmania and Trypanosoma to metabolize the pyrazolopyrimidine nucleus as though it were a purine base.
