Two-Dimensional Polyacrylamide Gel Electrophoresis of Proteins

Abstract

O’Farrell1 introduced the first high-resolution method for analyzing heterogenous populations of proteins. Earlier attempts had met with only limited success. O’Farrell’s1 two-dimensional Polyacrylamide gel electrophoresis (20-PAGE) technique separates proteins according to isoelectric point in one dimension and molecular weight in the second dimension. The independence of these separation criteria for proteins contributes to the high resolving power of the technique. After minor modifications, O’Farrell’s1 technique was first used to analyze nervous system proteins in my laboratory.2–5
