On the Influence of Plasma Kallikrein Inhibitors and Antihistaminics on Formalin Edema of the Rat Paw (Prepared Discussion Remark)

Abstract

It was already shown by H. Kaller that the development of the rat paw edema eliceted by different kinds of inflammatory drugs is inhibited to some degree by Trasylol. Trasylol is known to inhibit many proteinases including plasma kallikrein. It is thought that in the course of the events which lead to the edema, plasma kallikrein is activated which in turn liberates bradykinin. Thus the inhibiting effect of Trasylol in the development of edema seems to be due to the inhibition of plasma kallikrein. We checked if thioglycolic acid which destroys plasma kallikrein rapidly, also inhibits the edema development. We studied this in the formalin edema of the rats paw. Formalin was injected deep in the paw. Simultanously inhibitors of plasma kallikrein, f.i. Trasylol intra peritoneally. The inhibiting effect of different amounts of Trasylol is shown on Fig.1, that of thioglycolic acid on Fig. 2.
