The Effect of Prostaglandins, Epinephrine and Aspirin on Cyclic AMP Phospho Diesterase Activity of Human Blood Platelets and Their Aggregation

Abstract

Kloeze (1967) reported that low concentrations of PGE(1) inhibited adenosine diphosphate (ADP)-induced platelet aggregation in plasma of pig, rat and man. Emmons et al (1967) observed that PGE(1) inhibited platelet aggregation induced by other physiological constituents: collagen, thrombin, norepinephrine and serotonin. The extreme potency of PGE(1) in preventing the aggregation phenomenon and its implication with the adenylate cyclic AMP system, as demonstrated by Butcher et al (1967), suggested that the prostaglandin effect might be mediated by cyclic AMP in platelets. Consequently, several laboratories at about the same time reported that PGE(1) increased the synthesis of cyclic AMP by platelet membrane fractions or lysates (Wolfe et al, 1969; Zieve et al, 1969; and Marquis et al, 1969) and by intact platelets (Salzman et al, 1969; Vigdahl et al, 1969).
