Abstract

Adenosine and the 5′-adenine nucleotides inhibit cholinergic neurotransmission in guinea-pig ileum through actions at presynaptic P1-purinoceptors (Paton et al., 1985). However, it was not possible to show whether the adenine nucleotides acted directly on the receptor or whether their actions were indirect, subsequent to hydrolysis of the nucleotides to adenosine. Activation of adenosine A2 receptors in VA13 fibroblasts by adenine nucleotides required conversion of the nucleotides to adenosine (Bruns, 1980). Moody et al. (1984) demonstrated breakdown of 5′-ATP by guinea-pig ileum and suggested that P1-purinoceptor stimulation by 5′-ATP is partly direct and partly by the degradation products 5′-AMP and adenosine.
