Plasmid Replication

DNA Synthesis

Part of the book series: NATO Advanced Study Institutes Series ((NSSA,volume 17))

Abstract

Bacterial plasmids offer many advantages for the study of DNA replication and its control. These extrachromosomal genetic elements have been discovered in over 40 different bacterial genera and they display a wide variety of phenotypic characteristics (1–4). Moreover, within an individual taxonomic grouping of bacteria, such as the Enterobacteriaceae, many different types of plasmids have been identified and characterized. These plasmids may differ in their molecular size, DNA base composition, and replication properties (1–7). As would be expected, larger plasmids are more diverse in their functions, since they carry a larger number of genes. There is recent evidence, however, that only a relatively small region of a plasmid specifies the genetic information for its autonomous replication (8–11).
