Abstract

We have a more detailed understanding of the mechanism of replication of the DNA of bacteriophage ØX174 than of any other organism, largely because of its small size and the concentrated efforts of a number of laboratories. The viral DNA is a circular, single-stranded DNA molecule of 5,375 nucleotides, and the sequence of these is known (Sanger et al., 1977). After infection of E.coli, the first event is the synthesis of a complementary strand to form the duplex parental replicative form (RF). This RF is transcribed and then replicated several times to form progeny RF molecules. Finally, synthesis of viral single-stranded DNA occurs and progeny virus particles are produced.
