Abstract

Recently we described a series of experiments by which the orientation of the E.coli polA cistron was established with respect to the linked genetic loci metE and rha (Kelley and Grindley, 1967A). It was possible to orient the cistron unambiguously because of the unique enzymic properties of the wild type polA gene product, DNA polymerase I, and the properties of the products of three allelic polA mutations, polA1,polA6 and polA107.
