The Plasma Kinin-Forming System

Abstract

Because the nonapeptide bradykinin can elicit some of the phenomena associated with an inflammatory response, considerable attention has been devoted for some time to the plasma protein pathway leading to the generation of this mediator. Although earlier studies were devoted chiefly to in vivo models or to crude plasma fractions, recent work has focused on the use of highly purified proteins to examine the sequence and nature of their interactions in this pathway. From these investigations have come better physicochemical characterization of the components of the kinin-forming system, the recognition that several of these components in themselves have biological activity compatible with an inflammatory response, further examination of modes of activating the kinin-forming sequence, and an increased understanding of the multiplicity of interactions of this sequence with other plasma systems implicated in inflammation. This chapter will discuss information in these areas, with a focus on studies using human material.
