Abstract
It was Gaskell (1880) who originally made the observation that pH can have remarkable effects on cardiac contractility. Subsequently, although both the negative inotropic effect of acidosis and the positive inotropic effect of alkalosis on heart muscle have been well documented, we still do not fully understand the underlying mechanisms. There are several ways whereby protons might regulate resting and twitch tension. For example, the free sarcoplasmic Ca level could be changed, or the response of the contractile proteins to Ca might be altered. The latter possibility, namely a decrease of Ca sensitivity of the contractile proteins in acidosis, has been demonstrated by a number of workers in skinned preparations (e.g. Fabiato and Fabiato, 1978) as well as in isolated myofibrils from cardiac muscle (e.g. Schädler, 1967).
