Abstract
In choosing a method for subcellular fractionation, the investigator must first consider the particular question which he wishes to answer. In studies of the subcellular distribution of a substance of almost certain cytoplasmic localization, it is often sufficient to monitor (for purposes of estimating recoveries) the concentration of that substance in a crude “nuclear pellet” obtained by low-speed centrifugation of a sucrose homogenate. From most tissues, however, such a pellet is so heavily contaminated with cytoplasmic debris that it is totally unsatisfactory for situations in which any degree of nuclear localization is a serious possibility. For studies in which determination of nuclear localization of a substance is important, considerable effort must be given to the removal of cytoplasmic particles adhering to nuclei and to purifying the nuclei from such contamination.
