Developmental Toxicity and Nonhuman Primates

Interspecies Comparisons

  • Chapter
Issues and Reviews in Teratology

Abstract

A primary objective of experimental teratology is to determine potential adverse effects of chemicals and other environmental agents on development prior to widespread human exposure. Ethical considerations prohibit the testing of compounds for teratogenicity in the human population. This constraint forces us to make extrapolations of potential human risk from data obtained in laboratory animal species. Interspecies differences in teratologic sensitivity make the selection of an appropriate animal model a particularly challenging problem. The use of nonhuman primates has been advocated on the basis of their phylogenetic relatedness to humans, as reflected in anatomical, reproductive, and embryological similarities. One objective of this chapter is to examine results of developmental toxicity studies in nonhuman primates and determine how faithfully they match known effects in humans. Retrospective analyses like these are useful in validating animal models, but in order to enhance our predictive capability we need a better understanding of the nature and extent of interspecies variations in developmental toxicity.
