Abstract

Definition of the structure of primary transcription units of RNA and their corresponding translation units is crucial to an understanding of the organization and regulation of the eukaryotic genome. During the last decade, great progress was made in determination of the structure of transcription units in prokaryotes. However, the organization of transcription in eukaryotes appears to be more complex than that in prokaryotes. The rapid progress made in the analysis of the structure of the operon in bacteria depended on combined genetic and biochemical analyses. In eukaryotes, however, the most convenient systems for genetic studies (e.g., Drosophila) are often not readily amenable to conventional biochemical techniques. For this reason, the majority of the scientific effort has been directed toward other eukaryotic systems. This chapter is devoted mainly to a discussion of some aspects of transcription and translation in D. melanogaster and their relationship to the organization of the Drosophila genome. Particular emphasis is placed on certain molecular and electron microscopic properties of RNA transcription and maturation. The more general aspects of these topics will only be briefly covered here, as these have been dealt with either in other chapters of this handbook series or irr recent reviews.
