Cloning and Expression in Escherichia coli of Three Amylase Genes of a Strictly Anaerobic Thermophile, Dictyoglomus thermophilum, and Their Nucleotide Sequences

Abstract

In our search for microorganisms that produce heat-stable amylases, we have targeted extremely thermophilic microorganisms or caldo-active bacteria capable of growing at temperatures above 70°C, based on the concept that a thermophile produces thermostable enzymes. This screening led to a novel bacterium that we have named Dictyoglotnus thermophilum (Saiki et al., 1985), which produces an amylase complex with acidic pH optimum useful for industrial starch saccharification (Kobayashi et al., 1988). Difficulties encountered in purifying and characterizing each of the amylases in the complex prompted us to clone and express the amylase genes in Escherichia coli and then to examine their properties. This chapter briefly describes the taxonomic properties of D. thermophilum and the cloning of three amylase genes from this strain into E. coli.
