Abstract

Considerable progress has been made in developing therapeutic approaches to promoting axonal growth and remyelination in the adult mammalian CNS. It has proved possible, for example, to manipulate the cellular environment of lesion cavities or demyelinated areas of the CNS so as to promote axon regeneration and/or remyelination. Schwann cells are good promoters of axonal growth, and under certain conditions even astrocytes have been shown to possess growth-promoting properties (Ard and Bunge 1988; Ard et al. 1987; Bahr and Bunge 1989; Hatten et al. 1991; Kawaja and Gage 1991; Neugebauer et al. 1988; Silver 1988; Smith et al. 1986). Both Schwann cells and oligodendrocyte progenitor cells remyelinate axons when transplanted into the adult mammalian CNS (Duncan 1996). However, Schwann cells fail to become integrated into the CNS and astrocytes somehow perturb the neuron-Schwann cell interactions that lead to myelination (Franklin et al. 1992; Guenard et al. 1994).
