A Preliminary Report on the Cloning of a Constitutively Expressed Rat Liver Cytosolic ALDH cDNA by PCR

Abstract

Aldehyde dehydrogenase (ALDH, EC 1.2.1.3) is a ubiquitous enzyme that catalyzes the oxidation of acetaldehyde to acetic acid during ethanol metabolism. ALDH is located in virtually all mammalian tissues and found in all major subcellular organelles. The hepatic cDNAs for phenobarbital(PB)-inducible cytosolic ALDH, mitochondrial ALDH, microsomal ALDH, and tumor specific cytosolic ALDH have been cloned in the rat (Dunn, et al., 1989; Farres, et al., 1989; Miyauchi, et al., 1991; Jones, et al., 1988). However, the cDNA for a constitutively expressed rat liver cytosolic ALDH has not been cloned. While constitutively expressed cytosolic ALDHs have been characterized in the livers of numerous species, there is controversy in the literature as to whether this hepatic isoform of ALDH exists in the rat. To address this question, a PCR-based approach to cloning a constitutively expressed cytosolic ALDH from a rat liver cDNA library was employed.
