Abstract
Transformation of Populus has been reported previously for different species. However, most of the transformation procedures using disarmed Agrobacterium strains relied on regeneration of transformed shoots from callus (Tsai et al., 1994; Klopfenstein et al., 1991, Confalonieri et al., 1995). This has some disadvantages, since callus formation may lead to increased rate of somaclonal variation and regeneration of chimeric plants (Lee & Phillips, 1988). Here, we describe an efficient procedure for regeneration of transformed adventitious shoots directly from stem expiants, following inoculation with a disarmed A tumefaciens, strain EHA105. Three different selection schemes were assesed for improving transformation efficiency.
