Abstract
Since 1-nitropyrene (1-NP) is one of the most common nitrated poly-cyclic aromatic hydrocarbons (nitro-PAHs) found in the environment it has been studied as a model substance for mutagenic and carcinogenic nitro-PAHs affecting human health (Tokiwa and Ohnishi, 1986). 1-NP is metabolized by reductive and oxidative pathways (Djurić et al., 1986). Although a DNA adduct indicative of reductive metabolism, N-(deoxyguanosin-8-yl)-1-aminopyrene, has been identified in Salmonella typhimurium (Howard et al., 1983), in Chinese hamster ovary cells (Heflich et al., 1985) and in rats and mice (Stanton et al., 1985; Hashimoto and Shudo, 1985; Mitchell, 1988), it has been reported recently to be a minor adduct in rabbits and rats (Gallagher et al., 1988; Roy et al., 1989). Therefore, oxidative or oxidative-reductive metabolic pathways may be quantitatively more important in vivo.
