Analysis of Cytokine and Cytokine Receptor Production by Human Dendritic Cells
Dendritic cells (DC) have a unique ability to stimulate a primary T lymphocyte response. The presence of adhesion molecules on DC1 promotes the association of DC with T lymphocytes and the MHC antigen-peptide complex on DC triggers the responding T lymphocyte TCR (signal I). It is clear that additional cellular properties of the DC (signal II), as well as signalling via the adhesion molecules, are crucial to its function as a specialist antigen presenting cell (APC). 2 The nature of the DC co-stimulatory signal II remains unexplained but the possibility that DC produce cytokines such as IL-13 or IL-6, 4 which activate T lymphocytes, has been suggested. Convincing evidence that DC produce cytokines has not been forthcoming5,6 but it is clear that exogenous cytokines such as IL-17 and GM-CSF8 do impact on DC. These effects may be considered as a third signal involved in T cell activation (regulated by CD45) which primes DC to maximise their APC function.9
