Affinity labeling of dehydrogenases has been achieved with various reactive groups bound to nucleotide pyrophosphates. Bromoketones react preferentially with sulfhydryl-groups of the protein (Jörnvall, et al., 1975; von Bahr-Lindström, et al., 1985). In the case of lactate dehydrogenase modification, the small molecule 3-bromoacetylpyridine is attached to the imidazole part of an essential histidine (Berghäuser, et al., 1971).
