Abstract
Efforts to eradicate poliomyelitis encompass not only an appropriate vaccination policy but steps to verify the success of eradication campaigns as well. Conventional procedures for monitoring the circulation of virulent polio strains include the isolation of enteroviruses from sewage, their differentiation as polio or non-polio strains, and their intertypic and intratypic differentiation. We have developed a procedure which considerably facilitates this task: after a strain has been isolated and identified as polio, we extract its RNA, reverse transcribe it to cDNA, carry out PCR and analyze the amplified fragment with various restriction enzymes.
