Autocrine Control of Milk Secretion: From Concept to Application

Abstract

The rate of milk secretion is regulated acutely by frequency and completeness of milk removal from the mammary gland. Numerous experiments in dairy animals over more than 30 years (see Elliot, 1961; Linzell and Peaker, 1971) have shown that this acute regulation is exerted locally within each gland rather than by circulating galactopoietic hormones. Experiments in lactating goats also demonstrated that local regulation of milk secretion by milk removal was not related to physical distension of the gland by accumulated milk (Henderson and Peaker, 1984), but was, on the other hand, absolutely dependent on milk removal from the gland (Linzell and Peaker, 1971). This pointed to the involvement of a chemical mechanism, and suggested that local regulation of milk secretion was the result of changes in the degree of feedback inhibition exerted by a secreted milk constituent. Evidence for feedback inhibition of milk secretion is reviewed elsewhere in this volume (Peaker, 1995). Here we describe work which has identified and characterised the autocrine inhibitor in goat’s milk.
