Abstract

Milk is produced in the lactating mammary gland by the alveolar epithelium. Alveoli are composed of a single layer of secretory polarized cells that surround a lumen and interact both with one another and with their basement membrane. Caseins and other milk proteins are synthesized and secreted in response to lactogenic hormones. However, milk production also requires cell-cell and cell-extracellular matrix associations. In this article, I discuss how the use of culture models has unravelled different facets of the molecular control of mammary gland differentiation. I show that milk production is dependent on a specific component of the basement membrane and is under coordinate regulation by the matrix as well as hormones.
