A Novel Generation of Viral Vaccines Based on the ISCOM Matrix

Abstract

The outcome of the immune response to subunit antigen preparations will depend largely on the physical form in which the immunogens are presented to the immune system. Vaccines based on individual viral proteins or subunits consisting of one or more B- and T-cell epitopes in a monomeric form are often weakly immunogenic compared to live attenuated or inactivated whole virus vaccines (Morein et al., 1983; Morein and Simons, 1985). To enhance the immunogenicity of viral subunits, adjuvants are usually needed. Although most adjuvants facilitate a better induction of immunity than the antigen or subunit alone, resulting in higher titers of specific antibodies and T-helper (Th) cell responses, they may not induce cytotoxic T-cell (CTL) responses very efficiently.
