Abstract

Electrophoresis is a method for size-separation of charged polymers using electric field. A gel is a network of cross-linked polymers; see Figure 9.1. The principle of electrophoresis in a gel is very simple. When charged polymers are free to move in a gel, subject to an electric field, their mobilities within the gel are size-dependent. Consequently, as they have moved from one side of a container to another, they become separated according to their size. Electrophoresis is an important technique for DNA mapping: after cutting DNA molecules with enzymes, one can separate the various segments by electrophoresis.
