Abstract
It is now well established that a number of hormones synthesised in the hypothalamic area and carried to their adenohypophyseal site of action by a portal system, act to modulate the release of anterior pituitary hormones. Some of these hypothalamic regulating factors have now been characterised. The first to be demonstrated (1) was corticotrophin releasing factor (CRF); however, its structure is still not known. Thyrotrophin releasing hormone (TRH) was the first hypothalamic factor to be fully characterised, largely through the efforts of two groups led by Guillemin and Schally respectively (2,3) and shown to be a tripeptide (pyroglutamyl-histidyl-proline amide). The synthesis of TRH was closely followed by the identification and synthesis of gonadotrophin releasing hormone (GnRH), a decapeptide which stimulates the release of both LH and FSH.
