Abstract
The classic investigations of Sutherland and coworkers into the actions of glucagon and epinephrine on hepatic glycogenolysis led to the discovery of cyclic AMP and largely formed the basis for the second messenger hypothesis of hormone action. Figure 1 shows an early schematic representation of this hypothesis published by Sutherland and Robison in 1966. It is interesting to note that in addition to the adenylate cyclase system, the figure depicts a generalized scheme with a plasma membrane receptor X and an intracellular second messenger Y. Thus, the authors recognized the possible existence of alternative mechanisms for hormones acting through cyclic AMP.
